Yeah, I agree with that structure for 40 players, although you might want to consider starting the blinds at $100-$100 which will allow more manoeuvrability with the starting stack size.
Fielding 50 players would be more difficult, but I would suggest the following starting stack with blinds starting at $100-$100:
$100 x 7
$500 x 3
$1000 x 3
This would give each player $5,200 in starting chips. If colouring up were necessaru, you would have to change $1000 chips in for $100 simply because there won't be any $500s left. 35 yellows would be sufficient to colour-up the blacks.
Using this structure, there would be 650 chips in play initially but only 315 after the colour-up, totalling $260,000.
